In this state, PGC1α moves to the nucleus and acts as a transcription factor, increasing the expression of genes that code for proteins involved in fatty acid transport, fat oxidation, and oxidative phosphorylation. The activation by phosphorylation of PGC1α may occur in several ways involving AMPK, calcium calmodulin–dependent protein kinase, and p38 mitogen-activated protein kinase pathways. SIRT-1–mediated deacetylation also can cause activation. AMPK1 can work in two ways, either by activating PGC1α by phosphorylation or else directly by promoting the expression of enzymes involved in skeletal muscle oxidative effects and metabolism. This is supported by observations that, in obese subjects, the skeletal muscle is less oxidative and has, during fasting, lower AMPK activation (9). At the same time, AMPK activation also inhibits mammalian target of rapamycin (mTOR ) signaling, which is an important factor involved in regulating muscle mass (32).
